Though this variant occurs in the last exon of the gene and is not expected to result in nonsense mediated decay, it is predicted to cause loss of normal protein function through protein truncation. Other truncating variants in this final exon of BMPR1A have been reported in association with juvenile polyposis syndrome in ClinVar. This variant is not present in population databases (gnomAD). Therefore, this c.1533delA variant is classified as likely pathogenic.
