NM_014391.3(ANKRD1):c.37_93dup (p.Lys13_Gly31dup) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the ANKRD1 gene (transcript NM_014391.3) at coding-DNA position 37 through coding-DNA position 93, duplicating 57 bases. Submitter rationale: The c.37_93dup57 variant (also known as p.K13_G31dup), located in coding exon 2 of the ANKRD1 gene, results from an in-frame duplication of 57 nucleotides at nucleotide positions 37 to 93. This results in the duplication of 19 extra residues (KKNGNGEAGEFLPEDFRDG) between codons 13 and 31. This amino acid region is not well conserved in available vertebrate species. In addition, this variant is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
