Therefore, it has been classified as a Variant of Uncertain Significance. Experimental studies have shown that this missense change affects RYR1 function (PMID: 20142353). This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 1058 of the RYR1 protein (p.Glu1058Lys). This missense change has been observed in individual(s) with malignant hyperthermia susceptibility (PMID: 20142353). This variant is not present in population databases (gnomAD no frequency).
