Submitter rationale: This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 5352 of the TTN protein (p.Asp5352Asn). This variant also falls at the last nucleotide of exon 54, which is part of the consensus splice site for this exon. This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of autosomal recessive TTN-related conditions (external communication, internal data). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 978037). Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown.
