This variant lies in the POLE gene (transcript NM_006231.4) at coding-DNA position 5590, where A is replaced by T; at the protein level this means replaces isoleucine at residue 1864 with phenylalanine — a missense variant. Submitter rationale: Variant summary: POLE c.5590A>T (p.Ile1864Phe) results in a non-conservative amino acid change located in the DNA polymerase epsilon, catalytic subunit A, C-terminal domain (IPR013697) of the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251466 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance.
