Uncertain significance for Diamond-Blackfan anemia 1 — the classification assigned by Johns Hopkins Genomics, Johns Hopkins University to NM_001022.4(RPS19):c.393G>A (p.Leu131=), citing ACMG Guidelines, 2015: This RPS19 variant is rare (<0.1%) in a large population dataset (gnomAD: 1/251260 total alleles; 0.0004%; no homozygotes) and has not been reported in ClinVar nor the literature, to our knowledge. Bioinformatic analysis predicts that this synonymous variant would not affect normal exon 5 splicing, although this has not been confirmed experimentally to our knowledge. Due to insufficient evidence, we consider the clinical significance of c.393G>A to be uncertain at this time.
