NM_001244008.2(KIF1A):c.1208-40G>A was classified as Uncertain Significance by ARUP Laboratories, Molecular Genetics and Genomics, ARUP Laboratories, citing ARUP Molecular Germline Variant Investigation Process 2024: The KIF1A c.1243G>A; p.Gly415Ser variant (rs375833834), also known as c.1181-40G>A in transcript NM_004321.6, to our knowledge, is not reported in the medical literature or gene-specific databases. This variant is found in the African population with an overall allele frequency of 0.19% (46/24110 alleles) in the Genome Aggregation Database. The glycine at codon 415 is weakly conserved, but at least one computational algorithm (SIFT) predicts a damaging effect of this variant on protein structure/function. However, given the lack of clinical and functional data, the significance of the p.Gly415Ser variant is uncertain at this time.
