This variant lies in the DSP gene (transcript NM_004415.4) at coding-DNA position 995, where A is replaced by G; at the protein level this means replaces glutamic acid at residue 332 with glycine — a missense variant. Submitter rationale: DSP Glu332Gly has not been previously reported and is absent in the Genome Aggregation Database (http://gnomad.broadinstitute.org/). We identified this variant in a sudden death proband with no cause found on postmortem. The proband has no family history of an inherited heart disease or sudden death. In silico tools PolyPhen2, MutationTaster and CADD predict this variant to be deleterious however SIFT predicts this variant to be "tolerate". Based on this evidence we classify DSP Glu332Gly as a variant of 'uncertain significance'.
