Likely pathogenic for Pancytopenia due to IKZF1 mutations — the classification assigned by Johns Hopkins Genomics, Johns Hopkins University to NM_006060.6(IKZF1):c.548G>A (p.Arg183His), citing ACMG Guidelines, 2015: This IKZF1 variant is absent from a large population dataset, and has not be reported to ClinVar nor the literature to our knowledge. Three bioinformatic tools queried predict that this substitution would be damaging. The arginine residue at this position is evolutionarily conserved across all species assessed and is located in the third zinc finger (ZF3) of the Ikaros DNA-binding domain where other disease-associated variants have been identified. As this variant is apparently de novo and consistent with the patient's clinical findings, we consider c.548G>A to be likely pathogenic.
