Pathogenic for Neurodegeneration with brain iron accumulation 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001029896.2(WDR45):c.371_372del (p.Tyr124fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the WDR45 gene (transcript NM_001029896.2) at coding-DNA position 371 through coding-DNA position 372, deleting 2 bases; at the protein level this means shifts the reading frame starting at tyrosine residue 124, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This premature translational stop signal has been observed in individual(s) with clinical features of epileptic encephalopathy with cerebellar atrophy and/or nervous system abnormality (PMID: 26633542, 31665836). For these reasons, this variant has been classified as Pathogenic. ClinVar contains an entry for this variant (Variation ID: 976755). This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Tyr125Cysfs*13) in the WDR45 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in WDR45 are known to be pathogenic (PMID: 23176820, 24368176, 24621584, 25744623, 26790960, 27030146, 27652284, 28554332).
