This variant lies in the NAGLU gene (transcript NM_000263.4) at coding-DNA position 1843, where C is replaced by T; at the protein level this means replaces arginine at residue 615 with cysteine — a missense variant. Submitter rationale: This sequence change replaces arginine, which is basic and polar, with cysteine, which is neutral and slightly polar, at codon 615 of the NAGLU protein (p.Arg615Cys). This variant is present in population databases (rs199729746, gnomAD 0.1%). This missense change has been observed in individual(s) with clinical features of mucopolysaccharidosis type IIIB (PMID: 29606097). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. ClinVar contains an entry for this variant (Variation ID: 975752).
