Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000033.4(ABCD1):c.347G>A (p.Gly116Glu), citing LabCorp Variant Classification Summary - May 2015: Variant summary: ABCD1 c.347G>A (p.Gly116Glu) results in a non-conservative amino acid change located in the transmembrane domain (IPR011527) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 165658 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.347G>A has been reported in the literature in at least one Japanese individual affected with Adrenoleukodystrophy (Matsumoto_2003). These data however, do not allow any conclusion about variant significance. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. A different variant affecting the same codon (p.G116R) has been reported in patients in HGMD database. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as uncertain significance.
