Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001384910.1(GUCA1A):c.300T>A (p.Asp100Glu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces aspartic acid, which is acidic and polar, with glutamic acid, which is acidic and polar, at codon 100 of the GUCA1A protein (p.Asp100Glu). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with inherited retinal dystrophy (PMID: 19459154). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 974933). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive.
