NM_000243.3(MEFV):c.1920C>G (p.Ile640Met) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: MEFV c.1920C>G (p.Ile640Met) results in a conservative amino acid change located in the B30.2/SPRY domain (IPR001870) of the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be tolerated. The variant allele was found at a frequency of 2.4e-05 in 249068 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1920C>G has been observed in the hereozygous state, unknown phase with another MEFV variant, or in cis with a pathogenic MEFV variant in individual(s) affected with Familial Mediterranean Fever (Canete_2007, Accetturo_2020, Bustaffa_2025, Quiles_2025, Aksoy_2024). These data do not allow any conclusion about variant significance. Co-occurrences with other pathogenic variant(s) have been reported (MEFV c.1958G>A, R653H), providing supporting evidence for a benign role. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 31411330, 39042260, 40252570, 17665427, 40368691). ClinVar contains an entry for this variant (Variation ID: 97468). Based on the evidence outlined above, the variant was classified as VUS-possibly benign.
