Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000537.4(REN):c.58T>C (p.Cys20Arg), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine, which is neutral and slightly polar, with arginine, which is basic and polar, at codon 20 of the REN protein (p.Cys20Arg). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with autosomal dominant tubulointerstitial kidney disease (PMID: 21084044, 33532864). In at least one individual the variant was observed to be de novo. It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 974505). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. Experimental studies have shown that this missense change affects REN function (PMID: 21084044). For these reasons, this variant has been classified as Pathogenic.
