Pathogenic for Fanconi anemia — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000135.4(FANCA):c.3490C>T (p.Pro1164Ser), citing LabCorp Variant Classification Summary - May 2015: Variant summary: FANCA c.3490C>T (p.Pro1164Ser) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251432 control chromosomes. c.3490C>T has been reported in the literature in the presumed compound heterozygous, compound heterozygous, or homozygous state in multiple individuals affected with Fanconi Anemia (example, DeRocco_2014, Kimble_2018), including multiple individuals carrying a pathogenic variant in trans. These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
