Likely pathogenic for Mucopolysaccharidosis, MPS-III-B — the classification assigned by Human Genetics Unit, University Of Colombo to NM_000263.4(NAGLU):c.587C>T (p.Pro196Leu), citing ACMG Guidelines, 2015: The Pro196Leu variant in NAGLU gene has been reported in a Sri Lankan patient diagnosed with Sanfilippo syndrome type B. (PMID: 29881562), and was absent from large population studies. This variant causes non-conservative substitution of amino acids; i.e. substitution of Proline by Leucine may result in the alteration of the structure of the protein.
