The Leu608 residue is located in the KIX domain of CREBBP, a binding site for transcriptional activators (Wood et al. 2016). Based on the de novo origin of the variant and its rarity, the p.Leu608Pro variant is classified as likely pathogenic for Rubinstein-Taybi syndrome.
