This variant lies in the DST gene (transcript NM_001374736.1) at coding-DNA position 21500, where A is replaced by G; at the protein level this means replaces aspartic acid at residue 7167 with glycine — a missense variant. Submitter rationale: This sequence change replaces aspartic acid with glycine at codon 4544 of the DST protein (p.Asp4544Gly). The aspartic acid residue is moderately conserved and there is a moderate physicochemical difference between aspartic acid and glycine. The DST gene has multiple clinically relevant transcripts.
