Pathogenic for Von Hippel-Lindau syndrome; Chuvash polycythemia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000551.4(VHL):c.376_382delinsAA (p.Asp126fs), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the VHL gene (transcript NM_000551.4) at coding-DNA position 376 through coding-DNA position 382, replacing the reference sequence with AA; at the protein level this means shifts the reading frame starting at aspartic acid residue 126, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Asp126Asnfs*4) in the VHL gene. It is expected to result in an absent or disrupted protein product. For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in VHL are known to be pathogenic (PMID: 8956040, 12202531). This variant has not been reported in the literature in individuals with VHL-related conditions. This variant is not present in population databases (ExAC no frequency).
