Uncertain significance for Alstrom syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001378454.1(ALMS1):c.7816T>C (p.Ser2606Pro), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine with proline at codon 2607 of the ALMS1 protein (p.Ser2607Pro). The serine residue is moderately conserved and there is a moderate physicochemical difference between serine and proline. This variant is present in population databases (rs532551687, ExAC 0.03%). This variant has not been reported in the literature in individuals with ALMS1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Deleterious"; PolyPhen-2: "Not Available"; Align-GVGD: "Class C0". The serine amino acid residue is found in multiple mammalian species, suggesting that this missense change does not adversely affect protein function. These predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
