Submitter rationale: The p.E1071* pathogenic mutation (also known as c.3211G>T), located in coding exon 9 of the BRCA1 gene, results from a G to T substitution at nucleotide position 3211. This changes the amino acid from a glutamic acid to a stop codon within coding exon 9. This alteration was identified in 4 of 2168 Turkish individuals undergoing BRCA1/2 genetic testing based on a personal and/or family history of HBOC-related cancer (Bisgin A et al. Breast, 2022 Oct;65:15-22). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD).
