Uncertain significance for Neuropathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_013280.5(FLRT1):c.443C>G (p.Ala148Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine with glycine at codon 148 of the FLRT1 protein (p.Ala148Gly). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and glycine. This variant is present in population databases (rs80061941, ExAC 0.01%). This variant has not been reported in the literature in individuals with FLRT1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
