This variant lies in the RTEL1 gene (transcript NM_001283009.2) at coding-DNA position 3889, where C is replaced by T; at the protein level this means replaces proline at residue 1297 with serine — a missense variant. Submitter rationale: This sequence change replaces proline with serine at codon 1297 of the RTEL1 protein (p.Pro1297Ser). The proline residue is weakly conserved and there is a moderate physicochemical difference between proline and serine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with RTEL1-related conditions.
