This variant lies in the TTN gene (transcript NM_001267550.2) at coding-DNA position 2663 through coding-DNA position 2669, replacing the reference sequence with CTTCAC; at the protein level this means shifts the reading frame starting at aspartic acid residue 888, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This sequence change creates a premature translational stop signal (p.Asp888Alafs*11) in the TTN gene. While this is not anticipated to result in nonsense mediated decay, it is expected to create a truncated TTN protein. Information on the frequency of this variant in the gnomAD database is not available, as this variant may be reported differently in the database. This premature translational stop signal has been observed in individuals with autosomal dominant dilated cardiomyopathy (internal data).
