This variant lies in the DCTN1 gene (transcript NM_004082.5) at coding-DNA position 3034, where T is replaced by C; at the protein level this means replaces phenylalanine at residue 1012 with leucine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15"). This variant has not been reported in the literature in individuals with DCTN1-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces phenylalanine with leucine at codon 1012 of the DCTN1 protein (p.Phe1012Leu).
