NM_002439.5(MSH3):c.8G>A (p.Arg3His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R3H variant (also known as c.8G>A), located in coding exon 1 of the MSH3 gene, results from a G to A substitution at nucleotide position 8. The arginine at codon 3 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
