NM_007294.4(BRCA1):c.1015A>G (p.Lys339Glu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K339E variant (also known as c.1015A>G), located in coding exon 9 of the BRCA1 gene, results from an A to G substitution at nucleotide position 1015. The lysine at codon 339 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
