NM_000059.4(BRCA2):c.9651G>A (p.Met3217Ile) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 9651, where G is replaced by A; at the protein level this means replaces methionine at residue 3217 with isoleucine — a missense variant. Submitter rationale: Variant summary: BRCA2 c.9651G>A (p.Met3217Ile) results in a conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a benign effect of the variant on protein function. 4/4 computational tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 246540 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.9651G>A in individuals affected with Prostate Cancer and no experimental evidence demonstrating its impact on protein function have been reported. Three ClinVar submitters have assessed the variant since 2014: two classified the variant as uncertain significance, and one as likely benign. Based on the evidence outlined above, the variant was classified as uncertain significance.
