This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 9584, where C is replaced by T; at the protein level this means replaces threonine at residue 3195 with isoleucine — a missense variant. Submitter rationale: This missense variant replaces threonine with isoleucine at codon 3195 of the BRCA2 protein. This variant is also known as 9812C>T in the literature. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in individuals affected with breast or ovarian cancer (PMID: 18559594, 32846166). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
