NM_002528.7(NTHL1):c.538T>C (p.Tyr180His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y188H variant (also known as c.562T>C), located in coding exon 4 of the NTHL1 gene, results from a T to C substitution at nucleotide position 562. The tyrosine at codon 188 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear.
