Uncertain significance for Hyper-IgM syndrome type 5 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_080911.3(UNG):c.868T>G (p.Cys290Gly), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces cysteine with glycine at codon 290 of the UNG protein (p.Cys290Gly). The cysteine residue is highly conserved and there is a large physicochemical difference between cysteine and glycine. This variant is present in population databases (rs749817306, ExAC 0.001%). This variant has not been reported in the literature in individuals with UNG-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0").
