This variant lies in the PLEC gene (transcript NM_201384.3) at coding-DNA position 1589, where C is replaced by T; at the protein level this means replaces alanine at residue 530 with valine — a missense variant. Submitter rationale: This sequence change replaces alanine with valine at codon 557 of the PLEC protein (p.Ala557Val). The alanine residue is moderately conserved and there is a small physicochemical difference between alanine and valine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with PLEC-related conditions.
