NM_000282.4(PCCA):c.1275T>G (p.His425Gln) was classified as Uncertain significance for Propionic acidemia by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces histidine with glutamine at codon 425 of the PCCA protein (p.His425Gln). The histidine residue is moderately conserved and there is a small physicochemical difference between histidine and glutamine. This variant is present in population databases (rs548833871, ExAC 0.02%). This variant has not been reported in the literature in individuals affected with PCCA-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
