NM_000053.4(ATP7B):c.1351G>A (p.Gly451Ser) was classified as Uncertain significance for Wilson disease by Color Diagnostics, LLC DBA Color Health, citing ACMG Guidelines, 2015: This missense variant replaces glycine with serine at codon 451 of the ATP7B protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported with two pathogenic co-variants in an individual affected with Wilson disease, suggesting this variant may not contribute to disease (Qiao et al, 2021). This variant has been identified in 17/280504 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
