This variant lies in the CDKN2A gene (transcript NM_000077.5) at coding-DNA position 328, where T is replaced by A; at the protein level this means replaces tryptophan at residue 110 with arginine — a missense variant. Submitter rationale: The CDKN2A gene encodes two different proteins, p16INK4a and p14ARF, which are translated from alternative transcripts that have different open reading frames. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies.
