Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_002335.4(LRP5):c.1199C>A (p.Ala400Glu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine, which is neutral and non-polar, with glutamic acid, which is acidic and polar, at codon 400 of the LRP5 protein (p.Ala400Glu). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with autosomal dominant osteoporosis with familial exudative vitreoretinopathy and autosomal recessive osteoporosis pseudoglioma (PMID: 16252235, 25711638). ClinVar contains an entry for this variant (Variation ID: 967240). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt LRP5 protein function with a positive predictive value of 80%. For these reasons, this variant has been classified as Pathogenic.
