Uncertain significance for Pheochromocytoma/paraganglioma syndrome 3; Gastrointestinal stromal tumor — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003001.5(SDHC):c.11T>G (p.Leu4Arg), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SDHC gene (transcript NM_003001.5) at coding-DNA position 11, where T is replaced by G; at the protein level this means replaces leucine at residue 4 with arginine — a missense variant. Submitter rationale: This variant has not been reported in the literature in individuals with SDHC-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change replaces leucine with arginine at codon 4 of the SDHC protein (p.Leu4Arg). The leucine residue is moderately conserved and there is a moderate physicochemical difference between leucine and arginine. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
