Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001369.3(DNAH5):c.9626A>G (p.Lys3209Arg), citing Ambry Variant Classification Scheme 2023: The p.K3209R variant (also known as c.9626A>G), located in coding exon 57 of the DNAH5 gene, results from an A to G substitution at nucleotide position 9626. The lysine at codon 3209 is replaced by arginine, an amino acid with highly similar properties. This alteration was identified in a cohort of individuals undergoing whole exome sequencing for diverse clinical indications (Li AH et al. Genome Med, 2017 Oct;9:95). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear.
