Submitter rationale: This sequence change creates a premature translational stop signal (p.Ala301Profs*13) in the ALDH3A2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in ALDH3A2 are known to be pathogenic (PMID: 10577908, 10854114). In summary, this is a rare variant that is almost always observed as part of a complex European haplotype with a second pathogenic variant. It has been observed in several individuals with Sjögren-Larsson syndrome, and is expected to result in a disrupted protein product. For these reasons, this variant has been classified as Pathogenic.
