NM_004415.4(DSP):c.7657A>G (p.Ser2553Gly) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S2553G variant (also known as c.7657A>G), located in coding exon 24 of the DSP gene, results from an A to G substitution at nucleotide position 7657. The serine at codon 2553 is replaced by glycine, an amino acid with similar properties. This variant was reported in individual(s) with features consistent with dilated cardiomyopathy (Mazzarotto F et al. Circulation, 2020 Feb;141:387-398). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
