Uncertain significance for Temtamy preaxial brachydactyly syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014918.5(CHSY1):c.479A>T (p.Tyr160Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tyrosine with phenylalanine at codon 160 of the CHSY1 protein (p.Tyr160Phe). The tyrosine residue is highly conserved and there is a small physicochemical difference between tyrosine and phenylalanine. This variant is present in population databases (rs778545156, ExAC 0.2%). This variant has not been reported in the literature in individuals affected with CHSY1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
