NM_025099.6(CTC1):c.3280G>A (p.Ala1094Thr) was classified as Uncertain significance for Dyskeratosis congenita by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with CTC1-related conditions. This variant is present in population databases (rs773036519, ExAC 0.002%). This sequence change replaces alanine with threonine at codon 1094 of the CTC1 protein (p.Ala1094Thr). The alanine residue is highly conserved and there is a small physicochemical difference between alanine and threonine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
