NM_000238.4(KCNH2):c.3410C>A (p.Ser1137Tyr) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S1137Y variant (also known as c.3410C>A), located in coding exon 15 of the KCNH2 gene, results from a C to A substitution at nucleotide position 3410. The serine at codon 1137 is replaced by tyrosine, an amino acid with dissimilar properties, and is located in the C-terminal region of the protein. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
