Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies. This variant has not been reported in the literature in individuals with TSC1-related conditions. This variant is not present in population databases (ExAC no frequency). This sequence change results in a premature translational stop signal in the TSC1 gene (p.Glu1128Trpfs*7). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 37 amino acids of the TSC1 protein.
