NM_002109.6(HARS1):c.202C>T (p.Arg68Trp) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015. This variant lies in the HARS1 gene (transcript NM_002109.6) at coding-DNA position 202, where C is replaced by T; at the protein level this means replaces arginine at residue 68 with tryptophan — a missense variant. Submitter rationale: Variant summary: HARS c.202C>T (p.Arg68Trp) results in a non-conservative amino acid change located in the Aminoacyl-tRNA synthetase, class II domain (IPR006195) of the encoded protein sequence. Three of four in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 2e-05 in 251436 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.202C>T in individuals affected with Usher Syndrome and no experimental evidence demonstrating its impact on protein function have been reported.
