Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.5789C>T (p.Ser1930Leu), citing Ambry Variant Classification Scheme 2023: The p.S1930L variant (also known as c.5789C>T), located in coding exon 42 of the POLE gene, results from a C to T substitution at nucleotide position 5789. The serine at codon 1930 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
