This variant lies in the SGCG gene (transcript NM_000231.3) at coding-DNA position 730 through coding-DNA position 732, replacing the reference sequence with AGCAT; at the protein level this means shifts the reading frame starting at leucine residue 244, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This variant disrupts the C-terminus of the SGCG protein. Other variant(s) that disrupt this region (p.Thr251Serfs*29) have been determined to be pathogenic (Invitae). This suggests that variants that disrupt this region of the protein are likely to be causative of disease. This sequence change results in a premature translational stop signal in the SGCG gene (p.Leu244Serfs*37). While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 48 amino acids of the SGCG protein.
