Uncertain significance for Immunodeficiency 35 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003331.5(TYK2):c.149C>T (p.Ser50Leu), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine with leucine at codon 50 of the TYK2 protein (p.Ser50Leu). The serine residue is weakly conserved and there is a large physicochemical difference between serine and leucine. This variant is present in population databases (rs747484809, ExAC 0.009%). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0").
