Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000038.6(APC):c.856C>A (p.His286Asn), citing Ambry Variant Classification Scheme 2023: The p.H286N variant (also known as c.856C>A), located in coding exon 8 of the APC gene, results from a C to A substitution at nucleotide position 856. The histidine at codon 286 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, in silico predictors for this gene do not accurately predict pathogenicity. Based on the available evidence, the clinical significance of this variant remains unclear.
